目的:探讨摘除卵巢致肾虚骨质疏松症的病理机制,并研究补肾中药防治疗效及其作用机制。方法:摘除雌性大鼠双侧卵巢的方法复制肾虚骨质疏松症模型,采用具有补肾壮骨益精作用的补肾中药(高、低剂量)对实验大鼠治疗12周,以骨疏康颗粒剂、盖天力片作为阳性对照组,正常大鼠、假手术组作为标准对照组,模型大鼠作为空白对照组。用RT-PCR法检测肾虚骨质疏松症大鼠骨、肾、下丘脑组织中Smurf1/Smurf2的m RNA表达。结果:与模空组相比较,补肾中药高、低剂量组可不同程度上调骨、肾组织中Smurf1/Smurf2的m RNA表达水平(P〈0.01),并明显下调在下丘脑组织中Smurf1/Smurf2的m RNA表达水平(P〈0.01),其中,补肾中药高剂量组效果最佳(P〈0.05,P〈0.01)。结论:骨质疏松症的发生,可能与骨、肾、下丘脑组织中Smurf1/Smurf2的m RNA表达的异常变化有关;补肾中药通过调控骨、肾、下丘脑组织中Smurf1/Smurf2的m RNA表达,对于骨质疏松症有一定的防治作用。
Objective:To explore the etiological factors and pathology of the osteoporosis due to kidney deficiency by using ovariectomy,and to study the preventive and treatment effect of tonifying kidney Chinese medicine and its mechanism.Methods:By using ovariectomy,we established the rat model of osteoporosis due to kidney-deficiency.We treated the osteoporosis rat with tonifying kidney Chinese herbs(high dosage,small dosage) for 12 weeks.Simultaneously,Gushukang Granule and Gaitianli Tablets were used as positive control.Normal rat group and sham-operate group were set as standard control group,and model rats were set as blank control group.Rats in the normal group and model group were not given any treatment.RT-PCR method was used to investigate the expression of Smurf1/Smurf2 mRNA in the femur,kidney,hypothalamus of the rats.Results:Compared wiht the model and normal group,the tonifying kidney Chinese medicine high dosage,small dosage group could increased the expression of Smurf1/Smurf2 mRNA in femur and kidney(P〈0.01),and decreased the expression of Smurf1/Smurf2 mRNA in the hypothalamus(P〈0.01).The tonifying kidney Chinese medicine high dosage group's effect was best(P〈0.05,P〈0.01).Conclusion:The changing of SmurfI/Smurf2 mRNA expression in the femur,kidney,hypothalamus tissue,might be one of the important occurring mechanism of the osteoporosis due to kidney deficiency.The experimental Chinese herbs,which have the function of tonifying kidney,could adjust the expression level of the gene of Smurf1/Smurf2 in the femur,kidney and hypothalamus tissue,and that would be effective in preventing and treating to the osteoporosis due to kidney deficiency.
