采用PCR扩增和直接测序法分析了四川裂腹鱼乌江种群32个个体mtDNA控制区的序列差异和遗传多样性。在该种群mtDNA控制区长度为464bp的同源序列中,共计有9个变异位点,占总位点数的1.94%。32个个体共定义了9种单倍型,单倍型间平均遗传距离(P)为0.0060,单倍型多样度(Hd)为0.768。核苷酸多样性(π)为0.00324,平均核苷酸差异数(K)为1.500。用单倍型间遗传距离构建的NJ分子系统树聚为两个分支。结果提示四川裂腹鱼乌江种群遗传多样性匮乏,保护四川裂腹鱼乌江种群刻不容缓。
The genetic diversity and sequence variation of the mtDNA D-loop of 32 individuals from the population of Schizothorax kozlovi from the Wujiang River, Guizhou were studied via PCR and DNA sequencing. A total of 9 variable loci ( 1.94% of total loci) were detected in the 464 bp sequence of the mtDNA D-loop in the population. Nine haplotypes were identified from 32 individuals according to the determined sequences. The haplotype diversity (Hd) was 0.768, and the average genetic distance (P) among the haplotypes in the population was 0.0060. The nucleotide diversity (or) of the 32 individuals was 0. 00324, and the average number of nueleotide differences (K) was 1. 500. The NJ phylogenetic tree indicated that the 9 haplotypes from 32 Schizothorax kozlovi individuals were clustered into two clades. The genetic diversity of the population of Schizothorax kozlovi from the Wujiang River is very low and the protection for the population is necessary.
