目的:研究miR-34a在鼻息肉上皮细胞中的表达并探讨其对低氧诱导鼻黏膜上皮间质转化的影响。方法:利用实时荧光定量PCR检测鼻息肉和正常下鼻甲黏膜上皮细胞中miR-34a的表达情况;对正常鼻黏膜上皮细胞进行原代培养,并予低氧处理,通过Western-blot检测miR-34amimics转染对上皮间质转化相关蛋白(Ecadherin,α-SMA)表达的影响。结果:与正常下鼻甲相比,miR-34a在鼻息肉上皮细胞中表达减少,差异具有统计学意义(P〈0.05);低氧下调鼻黏膜上皮细胞中miR-34a表达,同时E-cadherin表达减少,α-SMA表达增加;这一效应能被miR-34amimics抑制。结论:miR-34a抑制低氧诱导的鼻黏膜上皮间质转化,可能在鼻息肉发生的病理生理过程中起重要作用。
Objective:To investigate the expression of miR-34 ain epithelial cells of nasal polyps(NPs)and to determine its effects on hypoxia-induced epithelial-mesenchymal transition(EMT).Method:Nasal epithelial cells(NECs)were collected from NPs and normal inferior turbinates.miR-34 aexpression was detected by means of quantitative reverse-transcriptase polymerase chain reaction.In addition,normal NECs were cultured under normoxia/hypoxia conditons.The effects of miR-34 amimics on hypoxia-induced EMT were investigated.Result:miR-34 awas specifically down-regulated in NECs of NP.Hypoxia decreased miR-34 aexpression in vitro.Furthermore,E-cadherin andα-SMA were down-regulated and up-regulated,respectively.miR-34 amimics transfection can reverse this process.Conclusion:miR-34 asuppress hypoxia-induced EMT in NECs.The down-regulation of miR-34 amay contribute to nasal polypogenesis.
