目的研究脂多糖(Lipopolysaccharide,LPS)诱导的急性肺损伤(ALI)小鼠模型中离体肺气体容积(excisedlunggasvolume,ELGV)和肺密度的变化规律,探讨ELGV和肺密度与动物模型肺形态学改变的相关性。方法90只健康雄性C57BL/6小鼠随机分为LPS4h组、LPS24h组和对照组,每组30只,LPS各组建立ALI模型。各组采用密度测量仪测量ELGV和肺组织密度,同时进行支气管肺泡灌洗液检查(BALF)、肺湿/干重(W/D)比值测定以及肺组织病理观察。结果急性肺损伤小鼠LPS4h组【(0.440±0.027)cm。】和24h组【(0.354±0.025)cm。】ELGV明显高于对照组【(0.217±0.011)cm3】,LPS24h组ELGV明显低于LPS4h组;LPS4h组【(O.363±0.030)g/cm3】和24h组[(0.497±0.038)g/cm。】肺密度明显低于对照组【(0.606±0.016)g/cm3】,LPS24h组肺密度明显高于LPS4h组(P均〈0.05)。结论ELGV和肺密度可以作为监测和评价急性肺损伤动物模型肺形态学改变的可行性指标。
Objective To study the changes of the excised lung gas volume (ELGV) and the lung density in the murine model with the acute lung injury (ALI) induced by lipopolysaccharide (LPS). in order to further clarify the relationships between the ELGV, the lung density and the lung morphometric alterations in animal model. Methods 90 healthy male C57BL/6 mice were randomly divided into three groups: LPS 4 h group, LPS 24 h group and a control group, 30 mice each group. The LPS treated groups of different time points served as the ALI models. A density determination kit was used to determine the levels of ELGV and lung density. Meanwhile, BALF, the lung wet/dry (W/D) weight ratio and pathological examination were assessed in each group. Results The ELGV values of both the LPS 4 h group [(0.440±0.027) cm3] and LPS 24 h group [(0.354±0.025) cm3] in the ALI mice were significantly higher than the control group [(0.217±0.011) cm3], and the ELGV value of the LPS 24 h group was significantly lower than the LPS 4 h group; while the lung density of both the LPS 4 h group [(0.363±0.030) g/cm3] and LPS 24 h group [(0.497±0.038) g/cm3] in the ALl mice were significantly lower than the control group [(0.606±0.016) g/cm3], and the lung density of the LPS 24 h group was significantly higher than the LPS 4 h group (P〈0.05). Conclusion The values of both the ELGV and lung density may be regarded as a potential index to monitor and assess the lung morphometric alterations in ALI animal model.
