目的:观察左归丸、右归丸对去卵巢骨质疏松症模型大鼠股骨骨髓组织Runx2、COLI基因与蛋白表达的影响,探讨左归丸、右归丸防治绝经后骨质疏松症的作用机制。方法:采用去卵巢方法建立绝经后骨质疏松大鼠模型,将60只SD雌性大鼠随机分为空白组、假手术组、模型组、补佳乐组、左归丸组、右归丸组,每组各10只。造模3天后灌胃给药12周,分别采用Q-PCR、Western blot法检测大鼠股骨骨髓组织中Runx2、COLI基因与蛋白的表达。结果:与空白组比较,模型组大鼠股骨骨髓组织Runx2、COLI基因与蛋白的表达均呈下降趋势(P〈0.01);与模型组比较,其余各用药组大鼠股骨骨髓组织Runx2、COLI基因与蛋白的表达均有不同程度的提高(P〈0.01或P〈0.05)。结论:左、右归丸可提高去卵巢大鼠股骨骨髓Runx2、COLI基因与蛋白的表达,达到促进骨形成的作用,其中左归丸的效果最佳。
Objective :To observe the effects of Zuogui Pill and Yougui Pill on ovariectomized osteoporosis model rats' Core Binding Factor Alphal 1 (Runx2), Collagen TypeI (COLI)gene and protein expressions in bone borrow and discuss the mechanism for the treatment of postmenopausal osteoporosis (PMOP). Methods:The postmenopausal osteoporosis rats model can be built by a- dopting the method of ovariectomy. Sixty SD female rats were randomly divided into the control blank group, the sham operation group, the model group, the progynova group, Zuogui Pill group, Yougui Pill group, each group ten only. Three days after building, use lavage for 12 weeks. The Q - PCR and Western blot method were used to detect the femur bone marrow tissue Runx2, COLI gene and protein expression. Results:Compared with control group ,model group rats' femoral bone marrow tissue Runx2 and COLI expressions were declined( P 〈 0.01 ). Compared with model group, the rest of the treatment group rats' femoral bone marrow tissue Runx2 and COLI expressions all had varying degrees of increase( P 〈 0. 01 or P 〈 0. 05 ). Conclusion : Zuogui Pill and Yougui Pill can improve ovariectomized rats' femoral bone marrow Runx2 and COLI gene and protein expressions to promote the role of bone formation. Zuogui pill's effect is the best.
