目的研究透明质酸酶(HAase)与基质金属蛋白酶-9(MMP-9)在人乳腺癌侵袭与转移中的意义。方法应用组织匀浆、酶连吸附测定法及免疫组化等方法测定HAase和MMP-9在人乳腺癌中的表达和其病理特征比较。结果①乳腺癌组织学分级Ⅰ、Ⅱ、Ⅲ级组HAase的分别为4.89±2.55、8.03±2.66、12.00±3.96mU/g(n=10,20,3),Ⅲ级组〉Ⅱ级组〉Ⅰ级组(P〈0.05)。乳腺癌有腋淋巴结转移组HAase表达(8.27±3.26mU/g,n=23)明显高于无腋淋巴结转移组(5.53±2.82mU/g,n=10)(t=-2.305,P=0.028)。②MMP-9阳性率在淋巴结转移组为(78.3%,18/23),明显高于无淋巴结转移组(30.0%,3/10)(P=0.016)。③MMP-9阳性组(n=21)HAase(8.57±3.12)mU/g明显高于阴性组(n=12)(5.47±2.85)mU/g(t=2.830,P=0.008)。结论HAase和MMP-9一样,与人乳腺癌侵袭与转移有关。
Objective To study the expression of hyaluronidase (HAase) and matrix metalloproteinase-9 (MMP-9 in the invasion and metastasis of human breast cancer. Methods The expression of HAase and MMP-9 in human breast cancer was detected using tissue homogenate preparing, enzyme-linked immunoadsorbent assay (ELSA), and immunohistochemistry examination. Results ① The levels of HAase were 4.89±2.55, 8.03±2.66, and 12.00±3.96 mU/g in grade Ⅰ, Ⅱ, and Ⅲ of breast cancer, respectively (n = 10, 20, 3), with significant differences between each other (P 〈 0.05). The HAase levels were significantly elevated in patients with axillary lymph node metastasis (8.27±3.26 mU/g, n = 23) as compared with patients without metastasis (5.53±2.82 mU/g, n = 10) (t = -2.305, P = 0.028). ② The positive rate of MMT-9 was significantly higher in patients with axillary lymph node metastasis (78.3%, 18/23) than patients without metastasis (30.0%, 3/10) (P = 0.016). ③ The expression of HAase was significantly greater in MMT-9 positive patients (8.57±3.12 mU/g, n = 21) than in MMT-9 negative patients (5.47±2.85 mU/g, n = 12) (t = 2.830, P = 0.008). Conclusions Both HAase and MMT-9 are associated with the invasion and metastasis of human breast cancer.
