为了研究钙调素基因对稻瘟病菌生长及致病性的调控作用,构建了稻瘟病CAMK基因敲除(同源重组替换)载体。Southern杂交分析表明,CAMK在稻瘟病菌基因组中为单拷贝。钙信号传导途径参与稻瘟病菌生长及致病性的调控,获得了潮霉素抗性转化子,转化子在菌丝生长、孢子萌发上与野生型无显著差异。
This paper studied the control role of calmodulin-dependent protein kinase (CAMK) in the growth and pathogenicity of M. oryzae and CAMK gene knockout (homologous recombination replacement) vector were constructed. The results showed that calcium signaling pathway was involved in the spore germination and hydrophobic conditions appressorium formation of the regulation. Meanwhile, Southern blotting analysis showed that CAMK gene exists as a single copy in M. oryzae genome. The transformant of hygromycin resistance was similar to the wild-type Guyll in continuous subculture of the hyphal growth characteristics and spore germination.
