目的探讨草苁蓉多糖(BRPS)对过氧化氢(H_2O_2)损伤的Hep G2细胞核转录因子(NF)-κB表达的影响。方法以Hep G2细胞为研究对象,通过H_2O_2诱导细胞氧化应激损伤模型,采用蛋白印迹法检测NF-κB、诱导型一氧化氮合酶(i NOS)、血红素氧合酶(HO)-1、环氧化酶(COX)-2以及核因子-E2相关因子2(Nrf2)的蛋白表达。结果 H_2O_2损伤Hep G2细胞后,NF-κB p65和Nrf2蛋白水平升高,i NOS、HO-1、COX-2蛋白水平升高。BRPS处理可降低Hep G2细胞NF-κB p65水平,对Nrf2蛋白水平无显著影响。BRPS同时降低i NOS、HO-1蛋白水平,但对COX-2蛋白水平无影响。结论 BRPS降低H_2O_2损伤的Hep G2细胞i NOS、HO-1蛋白水平,这可能与降低NF-κB活化有关。
Objective To study the effect of polysaccharides from Boschniakia rossica( BRPS) on expressiion of nuclear factor( NF)-κB in H_2O_2-damaged Hep G2 cells. Methods The protein expressions of NF-κB,inducible nitric oxide synthase( i NOS),heme oxygenase( HO)-1,cyclooxygenase( COX)-2 and nuclear factor erythroid2-related factor 2( Nrf2) were determined by Western blot. Results During the oxidative damage,nuclear levels of NF-κB p65 and Nrf2 proteins were increased,and the protein expressions of i NOS,HO-1 and COX-2 were up-regulated. The administration of BRPS reduced the nuclear NF-κB p65 level while there was no significant change of Nrf2 protein.BRPS also down-regulated the protein expressions of i NOS and HO-1,but had no significant effect on COX-2. Conclusions BRPS could reduce the levels of i NOS and HO-1 proteins,probably via the reduction of NF-κB activation.
