中药降乳散对大鼠催乳素瘤细胞表达Bcl-2、Bax的影响

目的研究中药降乳散对大鼠催乳素瘤细胞表达Bcl-2、Bax的影响。方法制备雌激素诱发大鼠催乳素瘤模型:成年雌性Wistar大鼠,切除卵巢,结扎子宫后随机分3组(n=5):对照组,皮下植入空白硅胶管 雌激素组,皮下植入含有己烯雌酚的硅胶管 降乳散组,皮下植入含有己烯雌酚的硅胶管,同时用中药降乳散汤剂(15mg·kg-1·d-1,指干燥中药的质量)灌胃。8周后处死动物,垂体称重,用放免法测定血清催乳素水平。Western blot检测凋亡相关蛋白Bax,Bcl-2在大鼠垂体组织中的表达。结果降乳散组的垂体质量、血清催乳素水平均明显低于雌激素组,具有显著差别(P〈0.01)。Bax在降乳散组的表达量明显高于雌激素组(P〈0.01)。Bcl-2在降乳散组的表达量明显低于雌激素组(P〈0.01)。结论中药降乳散具有促进催乳素瘤细胞凋亡的作用。本实验从分子水平探讨了中药抗催乳素瘤的机制,为中药治疗催乳素瘤提供了理论基础。

英文摘要:

OBJECTIVE To study the effect of Jiangru Power on the expression of Bcl-2 and Bax in rat prolactinoma. METHODS Adult Wistar rats were ovariectomized and divided into 3 groups at random. Each rat in control group (n=5) was subscutaneously implanted with a blank implant. Rats in estrogen group (n=5) were implanted with estrogen-containing implants. Rats in Jiangru Power group (n=5) were implanted with estrogen-containing implants, meanwhile, Jiangru Power were adminstrated (15 mg·kg-1·d-1) . After 8 weeks, all the rats were executed. The pituitary gland was weighed. Serum prolactin (PRL) levels were measured by RIA method. The expression of Bcl-2 and Bax in pituitary tissue was measured by Western blot method. RESULTS Pituitary weights, PRL levels in Jiangru Power group were lower than those in estrogen group (P〈0.001, respectively). The expression levels of Bcl-2 in Jiangru Power group were obviously lower than those in estrogen group (P〈0.001). The expression levels of Bax in Jiangru Power group were obviously greater than those in estrogen group(P〈0.001). CONCLUSION Jiangru Power may play a role in stimulating atoptosis in rat prolactinoma.
