目的:优选加味丹参饮抗大鼠心肌缺血再灌注损伤(IRI)的最佳配伍成分,探讨心肌IRI的中医治法。方法:采用结扎冠状动脉左前降支的方法制备心肌缺血再灌注(IR)模型,以乳酸脱氢酶(LDH)和肌酸激酶(CK)为指标,利用正交试验法,观察加味丹参饮不同配伍组方预处理对心肌IR后血清LDH、CK表达的影响。结果:与空白组比较,加味丹参饮能显著降低血清中LDH、CK的表达(P〈0.05);不同组方之间比较结果显示,加味丹参饮中丹参、黄芪、赤芍、红花4味中药作用最明显;比较各组极差发现,丹参对大鼠血清LDH、CK影响最大,其次为黄芪,再次为赤芍、红花。结论:加味丹参饮能显著降低IR大鼠模型血清中LDH、CK的表达,丹参、黄芪、赤芍、红花在本方组成中发挥主要作用,益气活血法能减轻心肌IRI。
Objective: To investigate the optimal formula of Jiawei Danshen decoction in the treatment of myocardial ischemia- reperfusion injury( IRI) in rats and the traditional Chinese medicine( TCM) treatment of myocardial IRI. Methods: The model of myocardial ischemia / reperfusion( IR) was established by ligation of the left anterior descending coronary artery. With lactate dehydrogenase( LDH) and creatine kinase( CK) as markers,an orthogonal test was designed to investigate the effect of pretreatment with various formulas of Jiawei Danshen decoction on the serum levels of LDH and CK. Results: Compared with the blank group,the Jiawei Danshen decoction group showed significant reductions in the serum levels of LDH and CK( P〈0. 05). The comparison of different formulas showed that Salvia miltiorrhiza,Astragalus membranaceus,Paeoniae Radix Rubra,and Carthamus tinctorius had the most significant effects. The comparison of range between each group showed that Salvia miltiorrhiza had the greatest effect on the serum levels of LDH and CK,followed by Astragalus membranaceus,Paeoniae Radix Rubra,and Carthamus tinctorius. Conclusion: Jiawei Danshen decoction can significantly reduce the serum levels of LDH and CK. Salvia miltiorrhiza,Astragalus membranaceus,Paeoniae Radix Rubra,and Carthamus tinctorius play important role in this formula,and the qi- tonifying and blood- activating method can alleviate myocardial IRI.
