目的探讨二氢卟吩e6(Ce6)光动力学疗法(PDT)对人结肠癌SW620细胞增殖、迁移及克隆形成能力等生物学行为的影响。方法 Ce6光动力处理SW620细胞,采用MTT法检测细胞增殖,划痕实验检测细胞迁移能力,平板克隆形成实验检测细胞克隆形成能力。结果 Ce6浓度和光照剂量增加时,Ce6-PDT对SW620细胞生长抑制作用逐渐增加,呈现浓度依赖、光照剂量依赖关系。划痕实验结果显示Ce6-PDT能够抑制细胞的迁移。Ce6-PDT能够减弱SW620细胞克隆形成能力。结论 Ce6-PDT能够抑制SW620细胞增殖、迁移及克隆形成能力。
Objective To explore the effects of chlorine e6-photodynamic therapy(Ce6-PDT) on proliferation,migration and colony formation of human colon cancer SW620 cells.Methods The inhibitory effect of Ce6-PDT on SW620 cell proliferation was detected by MTT assay.The migration and colony formation ability was determined by scratch test and colony formation assay respectively.Results Ce6-PDT inhibited the cell proliferation of SW620 in both concentration of Ce6-and dosage of irradiation-dependent manner.Results from scratch test showed that Ce6-PDT suppressed the migration of SW620 cells.The colony formation was decreased in SW620 cells after treatment with Ce6-PDT.Conclusion Ce6-PDT can inhibit the proliferation,migration and colony formation of human colon cancer SW620 cells in vitro.
