为了开发鸭坦布苏病毒间接ELISA抗体检测试剂盒,研究利用灭活纯化的鸭坦布苏病毒(DTMUV)FX2010株作为包被抗原,由瑞普(保定)生物药业有限公司连续制备了5批次鸭坦布苏病毒间接ELISA抗体检测试剂盒中试产品。该试剂盒对禽流感病毒、鸭瘟病毒、鸭细小病毒、I型鸭肝炎病毒、鸭呼肠孤病毒阳性血清和5份SPF阴性质控血清进行的检测均无交叉反应,表明具有较好的特异性。批内和批间重复性试验的变异系数为1.5%.4.2%和2.2%~5.3%,表明产品具有很好的稳定性。应用该试剂盒及中和试验对300份疑似DTMUV阳性血清样品进行检测,结果显示:二者阳性样品符合率为90.12%,阴性样品符合率为93.6%,总符合率为92.7%。研究结果表明,该试剂盒具有操作简便、敏感性高和特异性强的特点,可以为鸭坦布苏病毒病的快速诊断及其流行病学调查提供有力的工具。
In order to develop an indirect enzyme-linked immunosorbent assay (ELISA)kit for the detection of antibody against duck Tembusu virus (DTMUV), 5 batches of indirect ELISA antibody detection kits were produced by Ringpu (Baoding)biological pharmaceutical company limited using inactivated purified DTMUV (FX2010 strain)as coating antigen. The specific tests showed that there were no cross-reaction to the anti-sera against influenza virus, duck enteritis virus, duck parvovirus, duck hepatitis virus type I, duck reovirus and negative serums of 5 SPF duck. The inter-batch and intra-batch assay demonstrated that the coefficient of variation were 1.5% to 4.2% and 2.2% to 5.3%, respectively. A total of 300 suspected-positive DTMUV clinical serum samples were tested by indirect ELISA and neutralization test. The results showed that the positive and negative accordance rates were 90.12% and 93.6%, respectively, and the total accordance rate was 92.7%. Therefore, all results indicated that the kit used in this study was simple, sensitive and specific, and could be used for laboratory diagnosis and monitoring duck tembus virus infection.
