以75%乙醇为溶剂提取、聚酰胺型大孔树脂柱初步纯化,制得柿叶总黄酮;采用四氯化碳(CCl4)诱使昆明种小鼠急性肝损伤模型,研究柿叶总黄酮对肝脏的保护作用。结果显示,柿叶总黄酮能够明显减轻CCl4诱使的肝损伤,降低血清中ALT、AST活性和肝组织MDA含量,提高肝组织中SOD和CAT活性,且存在明显的剂量效应关系。上述结果表明柿叶总黄酮对CCl4所致急性肝损伤具有强的保护作用,作用机制可能与其对脂质氧化的抑制作用及其抗氧化活性有关。
The flavonoids were extracted from dried persimmon(Diospyros kaki L.f)leaves with75% ethanol and preliminarily purified with macroporous resin.The protective effect of persimmon leaves total flavonoids(PLTF)against carbon tetrachloride-induced(CCl4)on acute liver injury in rats of was investigated.The results showed that PLTF obviously reduced the liver damage,significantly decreased the AST and ALT levels in serum,remarkably lowered the liver indexes and MDA level,and elevated the SOD and CAT levels in hepatic tissues.These results indicate that PLTF has a protective effect against CCl4-induced acute hepatotoxicity in rats,and the hepatoprotective effects may be due to both the inhibition of lipid peroxidation and the increase of antioxidant activity.
