目的:观察益肺宣肺降浊方对血管性痴呆大鼠海马神经细胞凋亡和cAMP/PKA-CREB信号通路相关蛋白表达的影响以及探讨其部分作用机制。方法:80只健康SD大鼠随机分为假手术组、模型组、治疗组和对照组,每组20只。假手术组、模型组予盐水,治疗组予益肺宣肺降浊方,对照组予吡拉西坦灌胃。4组均采用Morris水迷宫实验检测大鼠记忆与空间探索能力,采用光镜和电镜观察海马神经元结构,Western blot检测海马PKA、P-CREB、BDNF、synapsin I蛋白表达水平。结果:与模型组比较,假手术组、治疗组及对照组逃避潜伏期缩短(P〈0.05);PKA、P-CREB、BDNF、synapsin I表达水平增高(P〈0.05);治疗组与对照组比较差异无统计学意义(P〉0.05)。结论:益肺宣肺降浊方可明显改善血管性痴呆大鼠学习记忆能力,减少神经细胞凋亡及提高海马PKA、P-CREB、BDNF、synapsin I蛋白表达水平,其作用机制之一可能是通过影响c AMP/PKA-CREB信号转导通路相关蛋白的表达来实现。
Objective: To observe the effects of Yifei Xuanfei Jiangzhuo Formula( YFXFF) on ultrastructure in hippocampus and cAMP / PKA-CREB signal pathways related to protein in rats with vascular dementia and to explore its part mechanism.Methods: Eighty SD rats were randomly divided into sham operation group,model group,treatment group and control group with 20 in each group. Sham operation group and model group were raised by normal saline and treatment group by YFXFF and control group by Piracetam. Four groups were tested with Morris water maze experiment testing memory of rats with space exploration ability and the ultrastructure in hippocampus in different groups were investigated by light and electron microscopy and PKA,P-CREB,BDNF,synapsin I by Western blot technique. Results: Compared with model group,sham group,the treatment group and control group's incubation period was shortened after treatment( P 〈0. 05). The contents of PKA,P-CREB,BDNF and synapsin I increased( P 〈0. 05). Compared with control group,it had no significance( P 〉0. 05). Conclusion: YFXFF can obviously improve the vascular dementia rats' learning and memory function,reduce the nerve cell apoptosis and increase the expressions of hippocampal PKA,P-CREB,BDNF and synapsin I. Its effect on the signal transduction pathway of cAMP / PKA-CREB may be one of the mechanisms.
