以刺槐的子叶为外植体在MS、LM 2种基本培养基上附加2,4-D、6-BA两种激素进行器官发生途径的研究,试验结果说明:基本培养基、激素种类和激素浓度对刺槐的植株再生有一定的影响;得出了刺槐器官发生途径植株再生的最佳培养基、愈伤诱导和分化的最佳激素浓度,刺槐诱导愈伤培养基最佳组合:MS+6-BA 1.0 mg·L^-1+2.4-D 5.0 mg·L^-1;刺槐愈伤分化培养基最佳组合:MS+6-BA 3.0 mg·L^-1+2.4-D 0.1mg·L^-1;刺槐不定芽根系诱导培养基最佳组合:MS+IAA 2.0 mg·L^-1+KT 1.0mg·L^-1.
In order to study organogenesis approach,explant of Robinia pseudoacacia L.was cultured in MS and LM medium in which two hormones(2,4-D and 6-BA) were added.The results showed that:(1)The basic medium,type of hormone and concentration of hormone have some effects on plantlet regeneration of R.pseudoacacia.(2)The best medium and concentration of hormone of callus introduction and differentiation for organogenesis and plantlet regeneration of R.pseudoacacia were found.The best medium of callus introduction are: MS+6-BA 1.0 mg·L^-1 + 2.4-D 5.0 mg·L^-1;the best medium of callus differentiation are MS+6-BA 3.0 mg·L^-1 + 2.4-D 0.1mg·L^-1;the best medium of adventitious bud and root introduction are MS+ IAA 2.0 mg·L^-1 + KT 1.0mg·L^-1.
