目的:探讨心肌缺血再灌注模型中细胞凋亡的发生及四逆汤的保护作用机制。方法:昆明种小鼠随机分为四逆汤组、模型组和正常组,观察心肌细胞 DNA ladder,检测超氧化物歧化酶活性、丙二醛含量、Bcl-2蛋白、Caspase-3活性和 Bcl-2、Caspase-3mRNA 转录情况。结果:模型组存在氧化损伤,出现 DNA lad-der,凋亡指数增加;四逆汤可减少凋亡对抗氧化损伤。模型组 Bcl-2蛋白表达增加,四逆汤组更为显著,与mRNA 转录结果一致;模型组明显升高的 Caspase-3活性在四逆汤组被显著抑制,mRNA 转录未见明显变化。结论:此缺血再灌注模型存在心肌细胞凋亡,四逆汤可能通过提高机体抗氧化能力,增加 Bcl-2蛋白表达,抑制 Caspase-3激活减少凋亡发生。
