目的通过对hsa-miR-197进行系统的生物信息学分析,预测其靶基因、相关功能及其可能参与的生物学过程。方法通过PubMed检索hsa-miR-197相关文献;通过miRBase、NCBI等在线数据库获取并分析hsa-miR-197序列保守性及其基因组特征。通过CA)功能注释、GO富集分析和信号转导通路富集分析,选择miRanda、TargetScan及PicTar预测hsa-miR-197的靶基因的交集作为分析的基因集合。结果(1)hsa-miR-197在人类同源的物种间具有高度保守性。(2)三种方法预测结果取交集获得hsa-miR-197可能的靶基因共有32个。(3)hsa-miR-197靶基因主要参与DNA修复、蛋白磷酸化、离子转运、蛋白修饰、DNA依赖的转录调控等生物学过程,涉及Wnt、氨基糖类代谢等信号转导通路。结论hsa-miR-197的功能较广泛,与发育、代谢等密切相关。
Objective To predict the targets,functions and biological process of hsa-miR-197 by bioinformatics analysis. Methods The hsa-miR-197-related data were searched by PubMed. The sequence and genome characteristics of hsa-miR-197 were analyzed on line by miRBase database and NCBI. miRanda, TargetScan and PicTar algorithm were used to predict the target genes of hsa-miR-197. The intersection of the three results as gene set was analyzed by GO annotation, GO overrepresentation (Gene Ontology) and pathway analysis. Results The hsa-miR-197 was highly conserved among the species of the human homolog. By searching for the literatures and finding the common target genes from miranda, TargetScan and PicTar, 32 predicted targets were obtained. GO analysis showed that the target genes of hsa-miR-197 might participate in the biological processes of DNA repair, protein phosphorylation, ion transport, protein binding, and regulation of transcription, DNA-dependent. The pathway analysis showed that these target genes were mainly involved in Wnt, aminosugars metabolism. Conclusion The target genes of hsa-miR-197 may have extensive functions and be closely related to the development, metabolism and etc.
