目的探讨血管内皮生长因子(vascular endothelial growth factor,VEGF)在体外对人急性白血病细胞株HL-607调亡的影响。方法采用形态学观察、单细胞凝胶电泳、流式细胞仪等方法对分别经过VEGF和(或)VP16处理过的急性白血病细胞株HL-60细胞凋亡进行检测。结果各种不同浓度VEGF(2μg/L、20μg/L、100μg/L)任孵育18h后可明显抑制急性白血病细胞株HL-60的凋亡,并且可对抗VP16诱导的细胞凋亡效应。结论VEGF可以抑制人急性白血病细胞的凋亡,可能是白血病的发病机制之。
Objective To investigate the effect of vascular endothelial growth factor (VEGF) on apoptosis of human acute myeloid leukemia HL-60 cell line in vitro. Methods The morphology of HL-60 cell and its apoptosis were detected by single-cell gel electrophoresis (SCGE) and flow cytometry (FCM). Resuits After treatment with VEGF(2μg/L、20μg/L、100μg/L) for 18 h, the apoptosis of HL-60 cells was inhibited in the way of dose dependence, and VEGF could also inhibite the apoptosis induced by Etoposide (VP-16) exposure. Conclusions The inhibitory effect of VEGF on apoptosis may play an important role in the pathogenesis of human acute myeloid leukemia.
