为了研究延边黄牛体细胞克隆胚胎内部的活性氧水平,以延边黄牛孤雌激活胚胎为对照,用2′,7′-二氯荧光素二乙酸酯和二氢乙锭两种染料分别对体细胞克隆胚胎进行染色,测定其内部超氧阴离子和过氧化氢含量。结果表明,2细胞期处理组间超氧阴离子水平差异不显著(P〉0.05),其它细胞期体细胞克隆组均显著高于孤雌激活组(P〈0.05),过氧化氢水平各细胞期体细胞克隆胚胎均显著高于孤雌激活胚胎(P〈0.05)。因此,延边黄牛体细胞克隆胚胎氧化损伤水平显著高于孤雌激活胚胎。
To study the intraeellular reactive oxygen species (ROS) level in somatic eell nuclear transfer in Yanbian Catlle, taking parthenogenetic activation embryos of Yanbian cattle as control, the cloned embryo somatic was dye by 2′, 7′- diehlorodihydrofluoreseein and hydroethidine to determine the internal superoxide anion and hydrogen peroxide content. The results showed that there was no significant difference in superoxide anion between two groups of 2-cell stage ( P 〉 0.05 ) , and the superoxide anion of reconstructed embryos was significantly higher than parthenogenetic activation embryos ( P 〈 0.05). The level of hydroethidine of reconstructed embryos was significantly higher than that of parthenogenetic activation embryos ( P 〈 0.05 ). The conclusions was drawn that oxidative damage level of reconstructed embryos in Yanbian cattle was significantly higher than that of parthenogenetic activation embryos.
