为了优化拟南芥悬浮细胞系的培养条件,以拟南芥愈伤组织为供试材料,研究了不同的接种量、蔗糖含量和转速对拟南芥悬浮细胞生长的影响情况。结果表明:接种量、蔗糖浓度和转速对拟南芥悬浮细胞的生长具有较大的影响,当接种量为4 g.150 mL-1时,悬浮细胞生长速度最快,鲜质量的增加量为0.463 g.mL-1;蔗糖质量浓度为30 g.L-1时细胞生长最好,干质量的增加量最高为0.198 g.mL-1;转速为120 r.min-1时,细胞增殖最快,能获得分散性与生长状况良好的细胞系。在优化后的培养条件下,拟南芥悬浮细胞呈现"S"型生长规律,继代周期为7 d。
In order to optimize culture conditions of suspension cell from Arabidopsis thaliana, taking callus in A. thaliana as experimental materials, effects of different inoculation quantity, sucrose concentration and rotation speed on growth of suspension cells were studied. The results show that inoculation quantity, sucrose concentration and rotation speed have significant influence on suspension cell growth from A. thaliana. The fresh mass of suspension cell is up to the highest value of 0.463 g · 150 mL^-1 when inoculation quantity is 4 g · 150 mL^-1. Cells grow well when sucrose concentration is 30 g · 150 mL^-1, and the highest dry mass increment of cell is 0.198 g · 150 mL^-1. Cell proliferation is the fastest when rotation speed is 120 r ·min^-1, and clone with good dispersity and growth status can be gained. Under the optimized culture condition, the growth ofA. thaliana suspension cells is "S" type, and subculture cycle of suspension cells is 7 days.
