以柔枝松的幼嫩顶芽为外植体,对不定芽的诱导、增殖和生根条件进行筛选。结果发现:在附加0.5mg/LBA和0.1mg/LNAA的SH培养基中,不定芽的诱导率为86.67%;诱导4周后,将外植体继代于SH基本培养基中,使不定芽伸长生长;然后,将伸长的不定芽转至含有2.0mg/LBA的SH培养基中进行增殖培养,4周后,增殖系数为3.75;继而在SH基本培养基或含有0.05%活性炭的SH培养基中进行壮苗培养;当不定芽长至1~1.5cm时,转入无植物生长调节剂的1/2GD培养基中进行生根诱导,培养6周后,在不定芽基部形成1~3条不定根;炼苗后,植株在土壤中(V(草炭土)∶V(蛭石)=1∶1)移栽成活率为64.86%。
The tender apical bud of limber pine was taken as explant to investigate the induction and multiplication of adventitious buds and the formation of adventitious roots.It was found that the induction frequency of adventitious buds was 86.67% when the tender apical bud was cultured on SH medium supplemented with 0.5 mg/L BA and 0.1 mg/L NAA.Four weeks later,the induced buds were subcultured on SH basal medium for elongation.Then the elongated shoots were transferred to SH medium containing 2.0 mg /L BA for multiplication for 4 weeks,and the number of average buds per shoot was 3.75.After cultured on the SH basal medium or supplemented with 0.05% AC,shoots with a height of 1-1.5 cm were transferred on 1/2 GD medium without growth regulator to induce roots.Six weeks later,1-3 adventitious roots were formed at the base part of adventitious buds.Finally,the plantlets were transplanted in soils(peat:vermiculite =1:1) and the survival rate was 64.86%.
