目的 探讨环氧合酶-2抑制剂(塞来昔布)对脑外伤大鼠大脑皮质水通道蛋白9(AQP9)表达的影响.方法 SD大鼠150只,随机分为假手术组、脑外伤组和塞来昔布组,各50只.采用Feeney's自由落体法制作脑外伤模型,采用干湿重法测定脑含水量,免疫组化和Western blot方法检测大鼠大脑皮质水通道蛋白9的表达.结果 脑外伤后3h含水量明显上升,6h时皮质含水量最高,24 h明显下降,而相同时间点的塞来昔布组大鼠含水量则明显较低(P<0.05).免疫组化和Western blot结果显示,相同时间点的脑外伤组大鼠大脑皮质AQP9蛋白表达量明显高于假手术组(P<0.05),而给予塞来昔布处理后AQP9蛋白的表达明显降低(P<0.05).结论 塞来昔布能够通过降低AQP9蛋白表达参与脑外伤水肿的调节.
Objective To study the effect of cyclooxygenase-2 inhibitor (celecoxib) on the expression of aquaporin-9 in the cortex of mice after traumatic brain injury.Methods Totals of 150 Sprague-Dawle (SD) rats were randomly divided into the sham group,traumatic brain injury group and celecoxib group (n =50 for each group).The traumatic brain injury model was developed according to Feeney's method.Dry-wet weight method was used to measure the extent of brain edema.The expression of aquaporin 9 in the cortex of rats was detected by immunohistochemistry and Western blot methods.Results Compared to the sham group,the brain water content was increased significantly at 3 h,to the peak at 6h,began to decrease at 24 h in the cortex of mice of traumatic brain injury (P < 0.05).Compared to traumatic brain injury group,the brain water content in the cortex of mice of celecoxib group at the same time point was decreased significantly.Immunohistochemistry and Western blot analysis showed that the expression of AQP9 protein was significantly higher in mice of the traumatic brain injury group than that in the sham group (P < 0.05).Compared to traumatic brain injury group,the expression of AQP9 protein in the cortex of the same time point in celecoxib group was decreased significantly.Conclusions Celecoxib could reduce edema of traumatic brain injury by reducing the expression of AQP9.
