目的探讨低强度超声对胶质瘤多药耐药蛋白三磷酸腺苷结合盒超家族G家族第2个成员(ABCG2)表达的影响。方法应用不同强度超声体外辐照胶质瘤(C6)细胞,MTT法检测C6细胞增殖活性,HE染色观察C6细胞形态。大鼠胶质瘤中,TUNEL法观察胶质瘤细胞的凋亡,免疫组化及Westernblot法检测ABCG2蛋白在胶质瘤细胞内表达的变化。结果低强度超声辐照后,c6细胞的增殖活性显著降低(P〈0.05)。大鼠胶质瘤内脑肿瘤组织小血管内红细胞渗出增加,胶质瘤细胞凋亡数显著增加(P〈0.05),ABCG2蛋白在胶质瘤细胞内表达显著下调(P〈0.05)。结论低强度超声通过降低C6细胞活性,诱导细胞凋亡和下调ABCG2表达,实现逆转多药耐药蛋白的表达。
Objective To investigate the effects of low-intensity ultrasound on the expression of multidrug resistant protein ATP-binding cas- sette super-family G member 2 (ABCG2) in glioma. Methods MTI? assay was applied to investigate the activity of glioma cells, and HE staining was adopted to observe the morphology of C6 cells in vitro. The apoptosis was detected by TUNEL assay, and the expressions of ABCG2 were determined by immunohistochemistry and Western blot assay. Results With the administration of low-intensity ultrasound, the activity of glioma cells was reduced, the apoptosis of glioma was increased, and the expressions of ABCG2 were down-regulated. Conclusion Low-intensity ultrasound could reverse the multidrug resistance of glioma by reducing the activity, inducing apoptosis and down-regulating the expression of ABCG2.
