为加强柞蚕核多角体病毒 (Altthraea pernyi nucleopolyhedrovirus, ApNPV )的分子基础研究,对ApNPV lef-12基因进行序列及转录分析。ApNPV lef-12基因长516个核苷酸,编码171个氨基酸,预测分子质量19.0kD。ApNPV lef-12基因5′端非编码区含有杆状病毒晚期基因启动子模序(ATAAG),但终止密码子(TAG)下游没有典型的多聚腺苷酸信号(AATAAA)。PSI-BLAST表明18种鳞翅目NPV编码ApNPV LEF-12同源蛋白:ApNPV LEF-12蛋白与类群INPV LEF-12的氨基酸一致性高于其与类群ⅡNPV LEF-12的氨基酸一致性。ApNPV LEF-12蛋白及与其关系较近的同源蛋白的有效密码子数较低。转录分析表明,ApNPV lef-12基因属晚期表达基因。
Antheraea pernyi nucleopolyhedrovirus (ApNPV) lef-12 gene is 516 nucleotides in length, which encodes a putative protein of 171 amino acids with a predicted molecular mass of 19.0 kD. The 5′-noncoding region contains one consensus promoter motif (ATAAG) for late gene transcription initiation. No typical putative polyA signal (AATAAA) was found at the downstream of the translational stop codon. PSI-BLAST search result revealed that 18 lepidopteran NPVs encode ApNPV LEF-12 protein homologues. The identity of ApNPV LEF-12 protein to that of group I NPV was higher than to that of group II NPV. The Effective Number of Codons (ENCs) of ApNPV LEF-12 protein and its closely related homologues were lower than that of the others. Reverse transcriptase (RT)-PCR analysis result indicated that ApNPV lef-12 was transcribed as a late gene.
