目的检测钙调蛋白在人自体中厚移植皮片中的表达,初步认识钙信号转导途径在自体移植皮片过度色素沉着发生机制中的作用。方法利用免疫组织化学方法,检测钙调蛋白在受区移植皮片、受区周围与供区周围正常皮肤中的表达并进行统计学分析。结果钙调蛋白的表达定位于表皮基底部黑素细胞、角质形成细胞的胞浆中;在自体移植中厚皮片组中,其表达的平均光密度值为0.49±0.08,显著高于受区周围正常皮肤对照组的平均光密度值0.23±0.08和原供区周围正常皮肤组的平均光密度值0.17±0.06,并与自体正常皮肤对照组中的表达差异有显著的统计学意义(P〈0.01);钙调蛋白在受区和供区周围正常皮肤对照组中的表达差异无统计学意义(P〉0.05)。结论钙调蛋白在自体中厚移植皮片中的表达较在自体正常对照皮肤组中显著提高,推测钙信号转导途径在自体移植皮片色素过度沉着中起着重要的调控作用。
Objective To investigate the expression of calmodulin in autoallergic intermediate thickness free skin graft and to recognize the role of calcium signal transduction pathway in hyperpigmented process of the skin grafts. Methods Immunohistoehemieal technique was adopted to detect the expression of calmodulin in the intermediate thickness free skin graft, the normal skin around the recipient area and normal skin separately and statistics was used to analyze the data. Results The expression of calmodulin was located in endochylema and cytolemma of melanocytes and keratinocytes in the basilar part of epidermis. The expression of calmodulin in most of intermediate thickness free skin graft was significantly higher than that in the control group. The positive integral optical density of calmodulin in the split-thickness skin grafts ( 0.49 ±0.08 ) was significantly higher than that in the normal skin around the recipient site (0.23 ±0. 08 ) and in the normal skin around the donor site (0.17 ±0.06), which showing statistically significant difference ( P 〈 0.01 ). The expression of calmodulin in the normal skin of the donor area was no statistic remarkably differences compared to the normal skin around the recipient area ( P 〉 0, 05). Conclusion The results indicate that the expression of calmodulin may markedly increase in intermediate thickness free skin graft and correlate with its pigmentation after the skin graft. Calcium signal transduetion pathway may play an important role in hyperpigmented process of the skin graft.
