内皮脂肪酶是1999年发现的,它与肝脂肪酶、脂蛋白脂肪酶和胰脂肪酶同属于三酰甘油酶家系。尽管这些不同的脂肪酶已被证实分别有着不同的特异性底物,但是序列分析显示内皮脂肪酶与脂蛋白脂肪酶有45%的同源性,与肝脂肪酶有着40%的同源性。自被发现以来,内皮脂肪酶一直就是研究的热门话题。许多研究结果显示内皮脂肪酶在血管内调节高密度脂蛋白方面扮演着重要的角色,同时它在某种程度上也参与了含apoB脂蛋白的代谢。另外,无论在体外实验还是体内实验,均显示内皮脂肪酶与胰岛素抵抗、炎症和高血压有着密切的关系。本综述的目的是综合近年来的研究结果,探讨内皮脂肪酶与代谢综合征的核心特征,即低高密度脂蛋白、高含apoB脂蛋白、胰岛素抵抗、肥胖、炎症和高血压之间的关系。

英文摘要:

Endothelial lipase , which was discovered in 1999, is belong to the large triglyeeridc lipasc gene family, including pancreatic llpasc , lipoprotein lipase(LPL) and bepatic lipase. Endothelial lipase exhibits a significant degree of homology with LPL(45% ) and hepatic lipase(40% ), although their substrate specificity have been shown to be quite different. Since its initial discoveu, endothelial lipase has been a topic of intense research. Studies have shown that it may play a key role in the intraxascutar remodeling of high density lipoprotein(HDL). It has also been suggested that endothelial lipase participants in the remodeling of an metabolism of apolipoprotein (apo)B-containing lipoproteins. Finally, endothelial lipase has also been been related to insulin resistance, obesity, inflammation, and hypertension in vitro as well as as in vivo. The purpose of this review is to discuss the association between endothelial lipase and some of the core features of the metabolic syndrome, namely low HDL-C, increased concentration of apoB-eontaining lipoproteins, insulin resistance, obesity, inflammation, and hypertension.
