目的观察siRNA下调Caveolin-1(Cav1)表达的效果及对体外培养C6胶质瘤细胞侵袭和迁移能力的影响。方法构建Cav1 siRNA质粒,转染体外培养C6胶质瘤细胞,利用Transwell体外侵袭实验和“划痕”实验分别检测细胞的侵袭和迁移能力,采用Western blot和RT—PCR的方法检测Cav1、EGFR、Pyk2的表达情况。结果所构建的Cav1 siRNA质粒转染体外培养C6胶质瘤细胞可明显下调Cav1的表达,干扰成功后C6胶质瘤细胞的侵袭能力和迁移能力明显下降,同时EGFR和Pyk2的表达明显减少。结论siRNA下调Cav1可降低体外培养C6胶质瘤细胞的侵袭和迁移能力。
Objective To study the in vitro migration and invasion of C6 glioma cell after Caveolin -1 (Car1) knocking- down by siRNA. Methods Cavl siRNA plasmids were transfected to C6 glioma cells. Abilities of cell invasion and migration were examined by Transwell and "Wound - healing" methods respectively. Expressions of caveolin - 1, EGFR, Pyk2 were measured by Western blot and RT - PCR. Results The siRNA plasmid used in this experiment can obviously knock - down the expression of Cavl. The invasion and the migration ability of C6 glioma cell decreased after siRNA as well as the expression of EGFR and Pyk2. Conclusions Cavl Knocking - down results in the decreasing of in vitro invasion and migration of C6 glioma. It maybe a potential target for anti - invasion therapy on gliomas.
