利用植物肌动蛋白保守片段设计简并引物,通过RT-PCR扩增出水葫芦肌动蛋白基因片段并连接到pMD19-T载体,转化E.coli,测序。序列分析结果表明,该序列与油棕、小野果蕉等植物的相似性程度达到85%以上,分子进化分析水葫芦肌动蛋白基因序列与油棕、小野果蕉等单子叶植物亲缘关系较近,但并没有与某些植物的肌动蛋白很好地聚成一类。本研究所克隆得到的水葫芦肌动蛋白基因序列能够为进一步研究水葫芦其他基因的表达提供内标。
Actin gene fragment from Eichhornia crassipes was amplified with degenerate primers designed according to the conservative sequences of plant actins by RT-PCT method. It was linked to pMD19-T vector and transformed to E.coli and then sequenced. Sequence analysis showed that the degree of similarity of this sequence and sequences from Elaeis guineensis and Musa acuminate etc. reached above 85%. Phylogenetic analysis also revealed that the sequence from E.crassipes was nearer relative to sequences form some monocot plants such as E.guineensis and M.acuminate etc., but it could not be clustered into one single group with some plant sequences. The sequence of this study can provide an internal control gene for further study of other gene expression of E.crassipes.
