目的探讨高尿酸对胰岛8细胞生存及功能的影响。方法采用免疫荧光染色方法检测正常小鼠(A组)和高尿酸小鼠(B组)胰岛形态及胰岛素水平变化。采用CCK-8方法观察尿酸浓度为0mg/dl(C组)、5mg/dl(D组)和15mg/dl(E组)大鼠胰岛细胞系INS-1细胞活力。采用ELISA法检测尿酸浓度为0mg/dl(F组)、15mg/dl(G组)小鼠分离的胰岛胰岛素分泌量及储藏量变化。结果与A组比较,B组小鼠胰岛面积明显缩小,胰岛素水平降低(P〈0.05)。与C组比较,D组、E组INS-1细胞活力呈剂量依赖性显著降低(P〈0.05)。与F组比较,G组细胞胰岛素的分泌水平和储存水平均显著降低(P〈0.05和P〈0.01)。结论尿酸对胰岛B细胞有直接损伤作用。
Objective To explore the effects of uric acid on the survival and function of pancreatic β-cells. Methods The changes in the size of the islets and insulin levels were detected by immunofluorescenee assay in mice with normal uric acid(group A) and those with hyperuricimic mice (group B). The activity of INS-1 ceils was observed by CCK-8 assay in the mice treated with different uric acid concentrations of 0 mg/dl(group C), 5 mg/dl(group D) and 15 mg/dl(group E). The changes of insulin secretion and storage were detected by ELISA in the mice treated with uric acid concentrations of 0 mg/dl(group F) and 15 mg/dl(group G). ResUlts The size of the islets in group B was smaller than that in group A and insulin level of group B was lower than that of group A (P〈0.05). Compared with group C, the activity of INS-1 ceils was decreased dosage-dependently (P〈0. 05) in groups of D and E. The insulin secretion and storage were lower in group G than those in group F(P〈0. 05). Conclusion High concentration of uric acid may directly damage pancreatic β-cells.
