从麻疯树基因组中克隆到一个CBF2基因(命名为JcCBF2).运用生物信息学的分析手段对该基因序列及其编码的蛋白质序列进行了分析,结果表明JcCBF2蛋白中不止包含保守的AP2/EREBP结合结构域,还具有CBF转录因子特征序列PKK/RPAGRxKFxETRHP、DSAWR.实时荧光定量PCR结果显示,JcCBF2基因主要在麻疯树叶片内转录表达,对低温胁迫极其敏感,但对干旱及盐胁迫不敏感,初步研究表明JcCBF2基因是低温诱导型转录因子.
A CBF2 gene was cloned from the Jatropha curcas genomes named JcCBF2. Bioinformatics a- nalysis was used for nucleotides sequences and encoded amino acid sequences of the JcCBF2. The results showed that the JcCBF2 contained the conserved AP2/EREBP DNA-binding domain. Besides, it also contained conserved motifs the PKK / RPAGRxKFxETRHP and the DSAWR, both of which were CBF signature sequences. The results of RT-qPCR showed that JcCBF2 was mainly expressed in leaves. The expression of JcCBF2 was induced by low temperature but not by dehydration or high salt, which re- vealed that the JcCBF2 was cold-inducible transcription factor.
