目的通过Ames试验与彷徨试验对蒲葵子正丁醇提取物的致突变试验结果,比较两种试验的灵敏度。方法对蒲葵子用正丁醇浸提,用Ames试验方法(掺入法)与彷徨试验方法,对TA100鼠伤寒沙门氏菌株进行致突变试验。结果两种试验方法结果显示,蒲葵子正丁醇提取物在5~500μg/ml剂量范围,以Ames试验方法与彷徨试验均检测出阳性结果,而在0.025μg/ml与0.5μg/ml剂量下,彷徨试验能检测出阳性结果,而Ames方法未检测出阳性结果。结论在较低浓度时,彷徨试验能检测出Ames试验检测不出的阳性结果,表明彷徨试验灵敏度较Ames试验高。
Objective To compare the sensitivity of the Fluctuation test and Ames test for examination of the sudden change caused by Chinese fan - palm sub - normal butyl alcohol extraction. Methods Chinese fan - palm was extracted with the normal butyl alcohol, and Ames test and Fluctuation test were carried out to detect TA100 mouse salmonella typhimurium. Results The results demonstrated that the extract in the range of 5 -500 μg/ml, the Ames test method and the fluctuation test method detected the positive result, and in the range of 0.025 - 0. 5 μg/ml, the Fluctuation test could examine positive result, but the Ames test could not change. Conclusion In low concentration, the Fluctuation test can examine positive result but Ames test can not, which indicated that the Fluctuation test has higher sensitivity than the Ames test.
