目的正交试验法优选天龙粉水溶性蛋白质的超声提取工艺。方法以牛血清蛋白标准液为标准品,紫外分光光度法测定吸光度,以水提液中蛋白质含量为考查指标,通过正交试验法探究料液比、超声提取时间、p H对提取效果的影响,从而优化提取工艺条件。结果在所选因素水平范围内,天龙粉水溶性蛋白质最佳提取工艺参数为:料液比为1∶40、超声提取时间为20 min、p H为9。在此条件下,天龙粉水溶性蛋白质的提取含量为50.789 mg/g。结论采用正交试验优化超声提取工艺,有效提高天龙粉水溶性蛋白质提取含量,具有较好的应用前景。
Objective: To optimize the ultrasonic extraction technology for water - soluble proteins from Gecko powder by orthogonal test. Method:Using bovine serum protein standard liquid as standard product, UV spectropho- tometry to determine absorbance, and the content of protein from water extract as the test index, through orthogonal test to explore the effect of the liquid ratio, ultrasonic extraction time, pH on extraction, and optimize extraction process conditions. Results:Within the scope of the selected factors level, the optimum extraction parameters of water - soluble proteins of Gecko powder were : the solid - liquid ratio was 1 :40, the extraction time of ultrasound was 20 min, and the pH was 9. In this condition, the extracting content of water - soluble proteins from Gecko powder is 50. 789 mg/g. Conclusion: Using orthogonal experiment to optimize ultrasonic extraction technology, effectively improve the extraction of water - soluble protein from Gecko powder, it has a good application foreground.
