Alzheimer＇s disease （AD） is affecting the health of more and more elder people all around the world. It is widely accepted that amyloid beta protein（Aβ） is the main cause of AD. As a crucial enzyme of Aft production, γ secretase determines the generation of different peptides of Aβ. In this review, recent advances of γ secretase modulators were summarized. Their structure and activity relationship was disclosed and future research directions were also discussed.
