脑胶质瘤是中枢神经系统最常见的恶性肿瘤。血肿瘤屏障(blood-tumorbarrier,BTB)的存在极大地限制了抗肿瘤药物进入脑肿瘤组织,降低了脑肿瘤的治疗效果,因此有效开放BTB成为提高脑胶质瘤化疗疗效的重要策略。近年来,越来越多的研究证实了多种转运药物到达肿瘤组织的方法,如脑室内给药、对流增强转运法、干扰BTB法以及选择性调控BTB的通透性等。其中,通过选择性调控BTB通透性,转运药物进入脑肿瘤组织具有效果好、副作用小的优点,是恶性脑肿瘤化疗中极具前景的治疗方法。本文概述了转运药物进入脑肿瘤组织的研究进展。
Gliomas are the most common malignancy of central nervous system.The existence of blood-tumor barrier(BTB)significantly limits the delivery of anti-cancer drugs into brain tumor,which reduces the therapeutic effect.Therefore,the opening of BTB effectively to improve the glioma chemotherapy has become an important strategy.In recent years,more and more studies confirmed various brain tumor drug delivery methods,such as intraventricular administration,convection enhanced transport,BTB interference and selective regulation of BTB permeability,etc.The methods of selective regulation of BTB permeability to deliver drug into brain tumor are the most promising pathway among chemotherapies of malignant brain tumors,which have a better prognosis with fewer side effects.This review outlines the research progress of brain tumor drug delivery.
