目的检测唾液腺腺样囊性癌(ACC)中瞬时受体势M(删)的表达及分布情况的变化,以探究ACE的发病机理。方法采用免疫印迹和免疫组织化学方法,检测TRPM在培养的ACC-2细胞及ACC组织中的表达,采用方差分析,利用Orig-inPro7.0软件进行统计学分析。结果ACC-2细胞和ACC组织中TRPM7蛋白的表达水平明显高于正常组织。TRPM7蛋白表达量经灰度值分析,正常腮腺组织为0.42±0.044,多形性腺瘤组织为0.413±0.085,而腺样囊性癌组织为0.85±0.045,差异有显著性意义(P〈0.001)。结论ACC中TRIM表达水平的增高可能是ACC发病的组织病理学基础。
Objective To investigate the expression and distribution of transient receptor potential canonical M(TRPM) in adenoid cystic carcinoma(ACC) of salivary glands, and evaluate the role of TRPM in the neural invasion of ACC. Methods The expression of TRPM in cultured ACC-2 cells and samples from normal salivary glands or ACC tissues were detected by Western blotting and immunohistochemistry. Resuits The enhanced expression of TRPM7 was observed in cultured ACC-2 cells and ACC tissues. Moreover, the enhanced TRPM7 expression was observed in the cytoplasm of ACC. Conclusion The enhanced density of TRPM expression in ACC may contribute to the etiopathogenisis of ACC.
