目的探讨疗癣卡西甫丸和银屑胶囊对咪喹莫特诱导银屑病小鼠模型皮损的影响。方法雄性BALB/c小鼠,随机分为正常对照组、模型对照组、阴性对照组、疗癣卡西甫丸组、银屑胶囊组以及阳性对照药物复方甘草酸苷组与地塞米松组,造模成功后予不同药物治疗2周,光镜下观察各组皮损组织形态学变化,计数炎症细胞浸润数、测量纵向表皮面积值;实时定量PCR方法检测皮损中TNF-α,IFN-γ,IL-17,IL-22细胞因子表达水平的变化。结果HE染色显示:与阴性对照组比较,疗癣卡西甫丸组、银屑胶囊组、复方甘草酸苷组、地塞米松组皮损厚度明显减小,表皮层较为平整,分别比较炎症细胞浸润数与纵向表皮面积值,差异有统计学意义(P〈0.05)。实时定量PCR检测结果显示,与阴性对照组相比,疗癣卡西甫丸组IFN-γ,IL-22表达水平,银屑胶囊组IL-17,IL-22表达水平,复方甘草酸苷组TNF-α,IFN-α,IL-17,IL-22表达水平,以及地塞米松组IL-17,IL-22表达水平均明显降低。结论疗癣卡西甫丸与银屑胶囊对咪喹莫特诱导的银屑病小鼠模型有明确疗效,可抑制在银屑病发病过程中起主要作用的Th1,Th17细胞通路。
Objective To observe the effects of Liaoxuan Kaxipu Pill and Yinxie Capsule in the treatment of psoriasis mice model induced by imiquimod. Methods Male BALB/c mice were divided into seven groups randomly : Normal control ( Norm. C ) , Model, Negative control ( Neg. C ), Liaoxuan Kaxipu Pill(LKP) , Yinxie Capsule (Y. Cap), Compound glycyrrhizin( Co. GH), and Dexamethasone (Dex) group. These groups were treated with different drugs respectively for two weeks. The histomorphology of mice skin lesions were evaluated according to the number of inflammatory cells and longitudinal epidermis area. Expressions of TNF-α, IFN-γ, IL-17 and IL-22 were investigated by real-time PCR. Results Compared with negative control, lesions of LKP-, Y. Cap-, Co. GH-, Dex-treated groups were thinner The number of inflammatory cells and longitudinal epidermis area of these 4 groups showed significant difference. Level of IFN-γ, IL-22 in LKP-group, IL-17, IL-22 in Y. Cap- and Dex-group, TNF-α, IFN-γ, IL-17 and IL-22 in Co. GH-group decreased significantly. Conlusion Liaoxuan Kaxipu Pill and Yinxie Capsule have definite therapeutic effect on imiquimod induced psoriasis mouse model by inhibiting Thl and Th17 pathway.
