目的:研究真姬菇多糖分子特性及生理活性,充分开发真姬菇资源。方法:沸水浸提真姬菇子实体多糖(HMP),用高效液相色谱(HPLC)及高效凝胶尺寸排阻色谱-十八角激光散色仪-示差检测仪联用(HPSEC-MALLS-RI)技术进行单糖含量和平均分子质量等分子特性的研究。选用WST-1法、Griess法和酶联免疫法(ELISA)研究HMP对人胃腺癌细胞(AGS)、结肠腺癌细胞(DLD-1)、子宫颈癌细胞(HeLa)的体外抑制作用及对小鼠巨噬细胞Raw264.7的免疫调节作用。结果:HMP由葡萄糖(89.2%)和半乳糖(10.8%)组成,平均分子质量分布于14770.0×104、1382.0×104、140.9×104、70.2×104u。HMP虽然对AGS、DLD-1、HeLa没有明显的体外抑制作用,但可以促进Raw264.7的增值和剂量依赖性地提高巨噬细胞中一氧化氮(NO)和前列腺素(PGE2)的生成量。结论:HMP具有免疫调节活性,可以作为免疫调节剂应用于保健品和医药行业。
For investigate the molecular characteristics and bioactivities of polysaccharides, HMP was isolated from Hypsizigus marmoreus fruit body by boiling water. Their monosaccharide levels included glucose content (89.2%) and galactose content (10.8%) obtained by HPLC system. The average molecular weight (Mw) of HMP was 14770x104, 1382x104, 141x104, 70x104 u determined by HPSEC-MALLS-RI system. Little cytotoxicity was apparent in cancer cell lines (AGS, DLD-1, HeLa) from the addition of HMP. However, HMP could enhance the proliferation of murine macrophage Raw264.7 cells and significantly stimulated Raw264.7 to release nitric oxide (NO) and prostaglandin E2(PGE2), suggesting their potential immunomodulating activities. The results of the current study demonstrate the possible applications of HMP as medicinal and functional food ingredients.
