大鼠为日本血吸虫的非适宜宿主,而小鼠为日本血吸虫的适宜宿主。前期研究表明细胞凋亡影响了日本血吸虫在不同适宜性宿主体内的生长发育,然而血吸虫中存在哪些凋亡相关基因及凋亡通路?哪些凋亡相关基因可能影响血吸虫的生长发育相关文献报道较少。本研究构建了日本血吸虫全基因组本地BLAST数据库,利用人的凋亡基因为索引搜索到了15个日本血吸虫凋亡相关基因。同时收集了大、小鼠来源32 d日本血吸虫,利用荧光定量PCR分析了32 d虫体中凋亡相关基因的表达情况,鉴定了大鼠和小鼠两种不同适宜性宿主来源32 d日本血吸虫差异表达的凋亡相关分子,为探讨日本血吸虫与不同适宜性宿主的相互作用机制提供了基础。
Schistosomajaponicum (S. japonicum) could infect more than 40 species of animals naturally, of which Mice are susceptible host to S. japonicum while rats are less-susceptible. Previous study have showed that apoptosis was one of the most important factors affect the development ofS. japonicum in different susceptible host. However, what genes involved in the apoptosis pathway of S, japonicum? Which apoptosis associated genes might affect the development ofS. japonicum. Still few study focused on these questions. The study built the whole genome BLAST database of S. japonicum firstly and dig out 15 apoptosis genes of S. japonicum using similarity search with human apoptosis genes. Meanwhile, 32 d S. japonicum worms from Wistar rats and BALB/c mice were collected separately. Real-time PCR analysis the expression level of these apoptosis associated genes in those worms. The results further identified the different expression apoptosis associate genes in S. japonicum worms from different susceptible host and provided the foundation for proceeding further research on parasite-host interaction.
