芋螺毒素(Conotoxins,CTxs)是一类特异作用于离子通道和膜受体的小分子多肽,是研究受体结构和功能及其相关疾病的重要工具。Mr IA是进入临床研究可用于镇痛治疗的一种T超家族的芋螺毒素。传统获取芋螺毒素是通过化学合成的方法,成本高、产量低。实验利用串联表达技术,构建原核表达载体,在大肠杆菌(Escherichia coli,E.coli)中成功表达了芋螺毒素Mr IA(recombinant Mr IA,r Mr IA)。通过溴化氰切割和纯化,最终1L菌液可获得纯度高达95%的r Mr IA30mg。小鼠热板实验表明,r Mr IA具有较好的镇痛活性。这为大量获得Mr IA以及其他芋螺毒素小肽的表达提供了方法。
CTxs are small peptides which specific target variety of voltage-gated ion channels and membrane receptor. Conotoxins are important tools for the study of receptors structure and function and their related diseases. Mr IA is a T-superfamily conotoxin which was entering clinical research for analgesia. The traditional methods to acquire conotoxin are chemical synthesis,while these methods are cost-consuming but low yields.Tandem repeats of mria gene fragment were constructed and ligated with prokaryotic expression vector. The r Mr IA was expressed successfully in E. coli. 95% purity r Mr IA were obtained by CNBr cleavage and purification,30 mg r Mr IA could be obtained from 1L media eventually. Mice hot plate test showed that r Mr IA had good analgesic activity. An effective method to obtain large quantities of Mr IA and a model for other small conotoxins expression were provided.
