目的:观察电针对慢性应激抑郁大鼠脑内ERK信号转导通路的影响,探讨针刺抗抑郁分子机制。方法:将32只SD大鼠随机分为空白组、模型组、电针组、百优解组,采用慢性应激结合孤养的方法造模。电针干预组选取"百会""印堂"穴,每次20min,每天1次,共治疗21d。百优解组给予百优解1.8mg/kg灌胃治疗,共21d。用Western blot方法检测海马中Ras、c-raf、p-c-raf、细胞外调节蛋白激酶(ERK)及p-ERK的含量。结果:大鼠海马中Ras、c-raf、p-c-raf、ERK含量各组之间没有显著性差异。海马中p-ERK水平模型组较空白组显著降低(P〈0.05),电针组与百优解组较模型组明显提高(P〈0.05)。结论:电针可提高抑郁大鼠脑组织内ERK信号转导通路的主要信号分子的磷酸化水平。这可能是针刺抗抑郁作用分子机制之一。
Objective: To observe the effects of electro-acupuncture (EA)on ERK signal pathway in the brain of depression rats model induced by chronic stress, and to explore the molecular mechanism of acupuncture in treating depression. Methods: 32 SD rats were randomly divided into 4 groups: control group, model group, ER group, and fluoxetine hydrochloride group. The method of chronic stress combined with solitary condition was adopted to make rats model. The rats of ER group were acupunctured on acupoints of Baihui (GV 20) and Yintang (EX-HN 3) with electro-acupuncture. The rats of fluoxetine hydrochloride group were gavaged with fluoxetine hydrochloride. Western blot was used to detect the level of Ras, c-raf, p-c- raf, ERK and p-ERK in the hippocampus of rats. Results: There was no significant difference among all the groups in the level of Ras, c-raf, p-c-raf and ERK. The level of p-ERK in the hippocampus in the model group was significantly lower than that in the control group (P〈0.05). Compare with model group, the level of p-ERK with the EA group and the fluoxetine group increased significantly (P〈0.05). Conclusion: EA could improve the phosphorylation of main signals in ERK pathway in the brain of depression model rats.
