目的研究B淋巴细胞瘤-2相结合抗凋亡基因I(BAGl)在外周神经系统损伤和修复中的作用。方法36只SD大鼠随机分为坐骨神经夹伤组(A组,28只)、假手术组(B组,4只)和正常对照组(C组,4只)。采用Westernblot检测BAGl和八聚体转录因子6(OCT6)蛋白的表达,免疫荧光染色观察BAGl与不同表型特异性标志物在细胞中的共定位情况。结果与B、C组相比,A组BAGl和OCT6蛋白表达增加(P〈0.01)。A组BAGl主要表达于Schwann细胞,并与OCT6、NF-200和S100有共定位。结论坐骨神经夹伤后BAGl表达上调,这可能与Schwann细胞分化相关。BAGl可能参与神经损伤后的髓鞘再生。
Objective To investigate the role of B cell lymphoma/leukemia-2-associated athanogene 1(BAG1) in the injury and repair of peripheral nervous system. Methods Thirty-six SD rats were randomly divided into three groups of A (sciatic nerve crush model, 28 rats), B (sham operation,4 rats) and C(normal controls,4 rats). The protein expressions of BAG1 and OCT6 were detected by Western blot, and the colocalization of BAG1 with different phenotypes of specific markers in the cells was observed by immunofluorescent staining. Results Compared with groups of B and C, the protein expressions of BAG1 and OCT6 were increased in group A(P〈0. 01). BAG1 was mainly expressed in Schwann cells and colocalized with OCT6, NF-200 and S100 in group A. Conclusion BAG1 expression is upregulated after sciatic nerve crush, which may be associated with the differentiation of Schwann cells. BAG1 may be involved in the remyelination process after nerve injury.
