目的:探讨真武汤对阿霉素所致小鼠肾损伤的保护作用及机制。方法:小鼠经尾静脉注射阿霉素10.5 mg/kg造模,实验分为正常组、模型组和真武汤组,每组12只,分组给药28 d。常规生化检测小鼠24 h尿蛋白总量(UTP)、血总蛋白(BTP)、血白蛋白(ALB)及血脂;电镜观察小鼠肾脏组织超微结构的变化;生化法检测小鼠肾组织SOD活力和MDA含量;Western Blot方法检测肾组织Nephrin和Podocin的表达水平。结果:模型组小鼠UTP、血胆固醇(TC)和三酰甘油(TG)显著增加,BTP和ALB显著降低;电镜下肾小球足细胞轻度肿胀,足突扁平、融合,足细胞、线粒体变性;肾组织SOD活力显著降低,MDA含量显著升高;肾组织Nephrin和Podocin蛋白表达显著降低。与模型组相比,真武汤组UTP、TC和TG均显著降低,ALB显著升高;肾脏超微结构变化明显减轻;肾组织SOD活性显著升高;肾组织Nephrin和Podocin蛋白表达显著升高。结论:真武汤能减轻阿霉素所致小鼠的肾损伤,其机制与减轻氧化应激损伤、维持nephrin、podocin的表达有关。
Objective: To investigate the effects of Zhenwu Tang( ZWT) on the expressions of nephrin and podocin in adriamycin nephropathy( MCN) mice. Methods: MCN model mice were established by a single intravenous injection of adriamycin were divided into model group and ZWT group,normal control group was set,each group contain 12 mice,they were given corresponding medicine or saline for 28 d The effects of ZWT on renal function,hemorrheologic parameters,rena1 ultmstucture,superoxide dismutase( SOD) and malondialdehyde( MDA) were observed. Western Blot was used to observe the expressions of nephrin and podocin in renal tissue. Results: Compared with normal group,the parameters of total proteinuria in 24 h,TC,TG and MDA were significantly higher,the BTP and ALB were significantly lower in the model group; the pathologic changes of renal ultrastructure in MCN mice showed podocyte flat and fusion; the protein expression levels of nephrin and podocin were markedly decreased. After ZWT treatment,the renal function and hemorrheologic parameters were significantly alleviated,the changes of ultmstucture,SOD and MDA were obviously ameliorated,the protein expressions of nephrin and podocin were up- regulated markedly compared with model group. Conclusion: ZWT might protect mice kidney from injury induced by ADR via increasing the expression of nephrin and podocin in podocytes.
