骨髓间充质干细胞对5/6肾切除大鼠肾脏损伤时Bcl-2、Bax表达的影响

目的探讨骨髓间充质干细胞(BMSCs)移植对5/6肾切除大鼠肾损伤的保护作用,观察BMSCs对肾组织Bcl-2、Bax表达的影响。方法将雄性SD大鼠BMSCs用PKH26标记后注入受体雌性SD大鼠体内。40只受体雌性SD大鼠随机分为4组(n=10):假手术组、5/6肾切除+DMEM-F12组(模型组)、5/6肾切除+贝那普利组(对照组)、5/6肾切除+BMSCs组(干细胞组)。60d后观察肾功能改变;免疫组化法检测Bcl-2和Bax的表达,并观察PKH26标记的BMSCs在受体大鼠肾脏的分布情况。结果假手术组肾功能明显好于其余各组(P〈0.05),对照组和干细胞组肾功能明显好于模型组(P〈0.05);模型组Bcl-2、Bax表达及Bcl-2/Bax比值明显高于其余各组(P〈0.05),干细胞组Bcl-2和Bax表达明显高于对照组(P〈0.05)。5/6肾切除后第60d可见标记的BMSCs定位于肾组织中。结论外源性BMSCs可减少Bcl-2和Bax蛋白表达,下调Bcl-2/Bax比值,减少肾组织细胞凋亡,从而有利于肾功能的早期恢复。

英文摘要:

Objective To study the protective effects of bone marrow mesenchymal stem cells(BMSCs) on 5/6 nephrectomy rats as well as observe the expressions of Bcl-2 and Bax proteins in the progression of renal injury.Methods BMSCs were isolated from male SD rats and labeled with PKH26 before injection.Totally 40 female recipient SD rats were divided randomly into four groups: sham group,DMEM-F12 infusion group(model group),benazepril gastric instillation group(control group) and BMSCs injection group(BMSCs group),each having 10 rats.After 60 days,the serum levels of BUN and SCr were observed.The expressions of Bcl-2 and Bax proteins were measured by immunohistochemical technique.The distribution of PKH26 positive BMSCs in the kidney of recipient SD rats was examined by immunohistochemistry.Results The renal function in sham group was significantly better than that in any other groups(P〈0.05);both control group and BMSCs group were significantly better than model group(P〈0.05).Bcl-2 and Bax expressions and the ratio of Bcl-2/Bax were higher in model group than in the other groups(P〈0.05);the expressions of Bcl-2 and Bax were higher in BMSCs group than in control group(P〈0.05).On the 60th day after 5/6 nephrectomy,BMSCs were found in renal tissues of BMSCs treated rats.Conclusion Exogenic BMSCs can decrease the expressions of Bcl-2 and Bax,lower the ratio of Bcl-2/Bax,reduce kidney cell apoptosis,and contribute to repairing the injured kidney.
