目的:研究血脂异常对ApoE基因缺失(apolipoprotein E-deficient,ApoE-/-)小鼠视网膜及Bruch膜的影响。方法:随机将24只2月龄(ApoE-/-)小鼠和24只C57BL/6J小鼠分别分为高脂组和普食组,每组动物12只,喂养4mo后检测其体质量、血浆总胆固醇,视网膜及Bruch膜行光镜和电镜检查。结果:6月龄ApoE-/-高脂组的血浆总胆固醇均显著性高于对照组(P〈0.05);6月龄ApoE-/-高脂组外核层厚度、色素上皮细胞层厚度显著低于对照组(P〈0.05),而其Bruch膜厚度则显著大于对照组(P〈0.05)。结论:血脂异常使ApoE-/-动物的视网膜光感受器细胞、色素上皮细胞及Bruch膜发生病理改变,其改变相似于早期年龄相关性黄斑变性的改变。
AIM:To research retina and Bruch's membrane of apolipoprotein E-deficient(ApoE-/-) mice effected by dyslipidemia.METHODS:Twenty-four 2-month-old ApoE-/-mice and 24 C57BL mice were randomly divided into group of normal chow and group of high fat chow,12 mice each group.Animals were killed four months later and plasma total cholesterol(TG) levels were determined.Light microscopy and transmission electron microscopy were used to observe retina and Bruch membrane.RESULTS:The plasma TG of the group of 6-month-old ApoE-/-mice high fat chow(HF) was significantly higher than that of control group(P0.05).The thickness of outer nuclear layer(ONL)and retinal pigment epithelium(RPE) was statistically lower than that of control group(P0.05),while the thickness of Bruch membrane was significantly higher than the control group(P0.05).CONCLUSION:The pathology change of photoreceptor,RPE and Bruch membrane was similar to the change of early age-related macular degeneration(AMD).
