目的:研究芍药苷对辐射致血虚证小鼠骨髓细胞IL-6、IL-18、IL-4R和IL-7R基因表达的影响,为阐明芍药苷补血作用的分子机制提供依据。方法:采用3.5 Gy60Coγ射线全身1次性照射制备小鼠血虚证模型,芍药苷按26.5 mg/kg.d的剂量连续灌胃给药7天;骨髓集落培养计数CFU-GM、CFU-E、BFU-E、CFU-m ix;RT-PCR检测骨髓IL-6、IL-18、IL-4R和IL-7R mRNA水平。结果:芍药苷显著升高血虚证小鼠骨髓CFU-GM、CFU-E、BFU-E、CFU-m ix数量,使其分别由7.50±3.11,71.00±11.46,8.50±1.29和7.25±2.22升至16.75±0.96,153.00±6.58,50.25±12.20和22.00±6.16;上调骨髓IL-6、IL-4R和IL-7R mRNA水平,使其与内参照的比值分别由0.26±0.02,0.19±0.04和0.11±0.02升至0.35±0.04,0.32±0.05和0.17±0.03,对IL-18 mRNA水平无显著影响。结论:芍药苷能促进骨髓造血祖细胞的增殖,上调骨髓细胞IL-6、IL-4R和IL-7R的基因表达。
Objective:To observe the effect of paeoniflorin on IL-6,IL-18,IL-4R and IL-7R gene expression in bone marrow cell of Xue deficiency mice induced by radiation and to provide the foundation for explaining the blood enriching mechanism of paeoniflorin.Methods: The Xue deficiency mice were induced by using a single dose of 3.5 Gy 60Co γ radiation.Myeloid colony cell was cultured and the number of CFU-GM,CFU-E,BFU-E and CFU-mix were observed and counted.IL-6,IL-18,IL-4R and IL-7R mRNA in bone marrow were measured by RT-PCR.Results: Paeoniflorin can raise significantly the number of CFU-GM,CFU-E,BFU-E and CFU-mix in bone marrow of Xue deficiency mice and up-regulate IL-6,IL-4R and IL-7R gene expression of bone marrow cell,but it is not of significant effect on IL-18 mRNA level.Conclusion: Paeoniflorin can promote the proliferation of hematopoietic progenitor cells of bone marrow and up-regulate IL-6,IL-4R and IL-7R gene expression in bone marrow cells.
