目的:研究非选择性M受体拮抗剂东莨菪碱对吗啡诱导大鼠伏核多巴胺释放的影响。方法:通过对大鼠运动行为变化观察和采用清醒动物微透析取样技术,用液相色谱电化学器检测动态测定伏核细胞外多巴胺含量变化。结果:吗啡(10 mg/kg,i.p.)能显著增加伏核细胞外多巴胺含量,东莨菪碱自身对伏核多巴胺释放无影响但能增加自发活动量,东莨菪碱(1.2 mg/kg,i.p.)能减少吗啡引起伏核多巴胺细胞外浓度增加,同时降低吗啡诱导活动度增加。结论:东莨菪碱可通过抑制吗啡诱导大鼠伏核多巴胺释放对大鼠中脑多巴胺系统发挥调控作用。
AIM: To investigate the effect of nonselective muscarinic antagonist scopolamine on the morphine-induced dopamine release in rat nucleus accumbens. METHODS: The extracellular level of dopamine in the nucleus accumbens was measured in awake freely-moving rats by microdialysis coupled with HPLC and locomotor activity was monitored by behavior test. RESULTS: Morphine( 10 mg / kg i.p.) significantly increased efflux of dopamine in the nucleus accumbens. Scopolamine had no effect by itself on the release of dopamine in the nucleus accumbens but elevated locomotor activity. Scopolamine( 1. 2 mg / kg) pretreatment attenuated the ability of morphine to increase extracellular concentration of dopamine and inhibited morphine-induced locomotor activity. CONCLUSION: These results suggest that scopolamine can modulate the function of midbrain dopamine system by inhibiting morphine-induced dopamine release in the nucleus accumbens.
