目的探讨雌激素对C57BL/6小鼠主动脉中ZFP580基因表达的影响。方法建立假手术组、去势对照组(卵巢切除)、去势实验组(卵巢切除并腹腔内注射苯甲酸雌二醇)C57BL/6小鼠高脂血症模型并鉴定;提取C57BL/6小鼠主动脉总RNA,设计并合成ZFP580及β-actin基因的引物,利用半定量RT-PCR方法分析ZFP580的mRNA水平。结果成功构建去势C57BL/6小鼠高脂血症模型;去势对照组小鼠ZFP580基因表达水平与假手术组和实验组相比均下降(P〈0.05),而后两者表达水平比较无显著差异(P〉0.05)。结论小鼠在摘除卵巢失去雌激素的调控后,ZFP580基因表达下调,小鼠在去势后定期给予雌激素,ZFP580基因表达回复至假手术组水平,说明雌激素能够上调ZFP580基因表达。
Objective To investigate the effect of estrogen on ZFP580 gene expression in the aortic tissue of C57BL/6 mice. Methods The C57BL/6 mice were made hyperlipidemia model and divided into three groups: sham-ovariectomized, ovarieetomized and ovarieeto- mized with estrogen groups. Then the ZFP580 gene expressions were analyzed by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) in the three groups. Results Results The ratio of ZFP580 gene and 13-aetin gene was significantly lower in the ovarieeto- mized group than that of sham-ovarieetomized and ovariectomized with estrogen the other two groups ( P 〈 O. 05 ). While the ratio was similar between the sham-ovariectomized group and ovarieetomized with estrogen group. Conclusions Without estrogen ,ZFP580 gene expression in the ovarieetomized group are downregulated. The expression of ZFP580 gene in the ovariectomized with estrogen group restores to the level of that of the sham-ovariectomized group. Estrogen can increase the expression of ZFP580 gene.
