目的全面研究Mr9000人颗粒溶素(GLS)的生物学活性.为其作为生物治疗剂应用奠定基础。方法将含Mr9000GLS编码基因的pZM03质粒转染肝癌细胞株SMMC-7721,通过MTT实验、流式细胞仪、Hoechst染色法检测GLS的致肿瘤细胞凋亡活性;将pZM03转染已感染结核分枝杆菌毒力株H37Rv的RAW264.7细胞,96h后裂解细胞作细菌培养及菌落计数,检测GLS的细胞内直接杀菌活性。结果将pZM03质粒转染SMMC-7721细胞后,从多个角度检测到了GLS的致肿瘤细胞凋亡活性;转染荷菌的RAW264.7细胞96h后,菌落计数实验证明GLS具有一定的细胞内直接杀菌活性。结论pZM03质粒编码的Mr9000GLS具有天然GLS的杀瘤活性和一定的细胞内直接杀菌活性。
Objective To investigate the biological activity of Mr 9000 human granulysin for the application as a biological therapeutic agent. Methods The eukaryotie plasmid pZM03 was transfeeted into the SMMC-7721 (a cell line of human hepatoma) and the cell apoptosis was detected by MTT assay, flow cytometry and Hoeehst staining. The bactericidal activity of GLS was examined by counting the colonies of H37Rv survived in maerophages RAW 264.7 transfected with pZM03 at 96 h. Results The capability of GLS for inducing apoptosis in tumor cells was confirmed by MTT assay, flow eytometry and Hoeehst staining. The colony counts showed that GLS had some bactericidal activity for Mycobacterium tuberculosis. Conclusions The GLS expressed by maerophages transfeeted with the eukaryotie plasmid pZM03 does have the ability of inducing apoptosis in tumor cells and killing intraeellular Mycobacterium tuberculosis.
