以越橘果实转录组测序文库为基础,借助生物信息学方法对越橘PDR型转运蛋白基因进行鉴定,并利用实时荧光定量PCR技术对其在越橘不同器官和组织中的相对表达量进行分析。结果表明:越橘果实至少含有30个PDR转运蛋白基因;从越橘果皮和果肉差异表达文库中筛选出21个差异表达基因,且都是在果皮中上调表达的基因;通过qRT-PCR分析,发现21个PDR转运蛋白基因在越橘不同器官和组织中的表达具有组织特异性,其中有16个基因在根中表达量最高。
Bioinformatics was used to identify blueberry PDR transporters based on transcriptome sequencing library of blueberry fruit. And real-time quantitative PCR was performed to analyze the relative expression levels of PDR genes in different organs and tissues of blueberry. The results showed there were at least 30 PDR transporters in blueberry fruit. Twenty-one differentially expressed genes were selected from the differences in tag frequencies in the skin and pulp libraries. And they were up-regulated genes in the skins of blueberry. The qRT-PCR results demonstrated that expression of21 PDR transporters had tissue specificity in different organs and tissues of blueberry,among which expression levels of 16 genes were the highest in the roots.
