在生物信息学分析和克隆番茄组蛋白去乙酰化酶HD2亚家族的基础上,探究了HD2家族在番茄果实发育阶段的表达模式.结果显示,番茄HD2家族有3个成员,其氨基酸序N端具有典型的MEFWG五肽基序,中部为疏水结构域分隔开的两段富含酸性氨基酸的结构域,C端的序列呈多样性,并且具有C2H2型锌指结构;亚细胞定位预测发现该家族成员均定位于细胞核,这与其参与组蛋白修饰功能相一致.表达模式结果显示,HD2基因家族在果实发育阶段中具有阶段特异性,推测其在调控番茄的果实发育方面有重要作用.
Histone acetylation plays a key role in regulating chromatin organization and gene expression. In this study, we successfully identified and cloned three tomato HD2 genes, SIHDT1101, S1HDT1102 and S1HDT1103. All the three tomato HD2 proteins had a characteristic domain strueture-a conserved pentapeptide motif (MEFWG) on the N-terminal and two highly charged acidic stretches in the central part. In addition, both S1HD11101 and S1HDT1103 contained a C2H2 type zinc finger in the C terminal. Furthermore, all the three HD2 proteins were localized in the nuclear. The gene expression analysis showed that each of the three HD2 genes displayed a specific expression pattern in tomato fruit development, indicating their important functions during fruit development.
